Abstract:

Cancer causes about 13% of human deaths and has not yet been properly treated. Conventional cancer treatment METHODs such as surgery, chemotherapy, and radiotherapy have many side effects and in some cases have no effect on cancer treatment, so creating appropriate and targeted therapies can revolutionize cancer treatment. New METHODs include gene therapy and using immunotoxins, which can act up to a high percentage. This study aimed to investigate the effect of modified PA protein that binds only to cancer cells along with lethal factor (LF) on cancer cells. In this regard, recombinant MPA and LF proteins were isolated from recombinant Bacillus subtilis and after examining the presence of protein bands and their concentrations using SDS-PAGE and Bradford METHODs, respectively, the combined effect of different concentrations of these proteins on lung cancer cells, Breast, prostate and pancreas were examined by MTT METHOD. The results showed that 75 ng of recombinant LF protein and 50 ng of recombinant MPA protein had the greatest effect on cancer cell death and caused the death of more than 98% of the studied cancer cells. With these results, it is hoped that a new dimension can be added to cancer treatment by producing targeted toxins such as MPA and LF.

Abstract:

Objective: Smartphone is an important technology device in our lifestyle. It has an important part of our daily lives, but it also has a negative effect, such as cell phone dependency. This research aimed to evaluate the psychometric properties of mobile phone abuse (MPA) in the Iranian population. METHOD: In this study, data were chosen from 1100 participants who were studying in Tehran universities. The principal version of the scale was translated into Persian using the back translation METHOD. All attendees completed Demographic Questionnaire, MPA Questionnaire, and Mobile Phone Problematic Use Scale (MPPUS). Eventually, a clinical interview (based on the fifth version of DSM) was done for all the participants. For data analysis, internal and external consistency, factor analysis, construct validity and confirmatory factor analysis (CFA) were used. Statistically, less than 0. 05 were considered to be significant. Results: According to expert judgments, content validity index was satisfactory. Furthermore, the reliability of the questionnaire was confirmed with Cronbach's α of 0. 90 and test-retest reliability of 0. 56 after 3 weeks. The best cutoff point for this questionnaire (MPA) was 46. Also, 4 factors were extracted by principal components METHOD and varimax rotation: "excessive use of cell phone, " "addictive use of social networks, " “ mood modification, " and "preoccupation" for both male and female students. Conclusion: MPA could be used in studies on the evaluation of mobile phone addiction. This can be a stepping stone towards the identification of problems and improvement of students' mobile phone abuse.

Abstract:

Progestins are useful for hormonal prevention of pregnancy in humans and animals. Progestrone and medroxy progestrone acetate are used for treatment of premenopausal syndrome and decreasing complications of estrogen therapy after menopause. Prescription of progesterone in dogs is usually done to control or delay estrus. If this drug is used at anestrous it will inhibit the gonadotropin excretion beyond the basic level and it will prevent cycle from happening again in proestrus or at the beginning of estrus progesterone will prevent ovulation. Use of progesterone is not totally safe in bitches. It can cause several problems such as cystic endometrial hyperplasia and pyrometera. In this study the effects of medroxy progestrone acetat (MPA) on electrocardiographic changes are evaluated. After nutritional and environmental uniformity and other evaluations, the animals were divided into two groups with six bitches in each group. MPA was used at 10 mg/kg for the first group and 20 mg/kg for the second group. An ECG was recorded from the first group in time zero. MPA was injected intramuscularly and immediately after ECG recording and the time of injection was noted. Another ECG was recorded 0.5 and 2 hours after injection. The same process was repeated for the second group. It was observed that MPA at 20 mg/kg increased the heart rate and consequently PR interval and RR interval were decreased significantly in normal range. Also administration of drug at 10 mg/kg decreased the heart rate and increased PR interval, QT interval and RR interval significantly in normal range. It is probable that these observations are the result of MPAs dose dependent vasodilatory nature that induces the heart to exhibit compensative reactions such as increasing heart rate.

Abstract:

Mycophenolic acid (MPA) is a fungal metabolite possessing antiviral, antifungal, antibacterial, antitumor and anti-psoriasis activities. It is being used as an immunosuppressive agent in kidney, heart and liver transplantation patients. In the presence of MPA, the proliferation of the B and T lymphocytes is inhibited. The MPAB and MPAE genes reside in a 25 kb gene cluster in the genome of Penicillium brevicoMPActum. In this study, the genomic DNA was extracted from P. brevicoMPActum grown on potato dextrose (PD) medium. To amplify the MPAB and MPAE fragments, the specific primers were designed using Gene Runner software according to P. brevicoMPActum IBT23078 sequence database under HQ731031. 1 accession number. The amplified MPAB and MPAE genes were cloned in the PTG19-T PCR cloning vector and transformed to Escherichia coli (E. coli) top 10 competent cells. The insertion of MPAB and MPAE in the PTG19-T cloning vector was further confirmed by PCR. The MPAB and MPAE amplification produced amplicons of 1477 and 780 (nt), respectively, with the same length according to the MPAB and MPAE genes deposited in the GenBank. However, the alignment results showed some differences at nucleotide and amino acid levels, implying a new strain of P. brevicoMPActum.
